Cover: colocalization of endocytosed surfactant protein (SP) A and lipid and of early endosomal markers in type II cells by confocal laser scanning microscopy. Top left: type II cells were incubated with unlabeled SP-A and rhodamine-phosphatidylethanolamine-labeled liposomes (red) for 3 min at 37°C and then incubated at 3°C for 30 min with FITC-labeled concanavalin A to visualize the cell boundaries, demonstrating true internalization of the lipid. Top right: to demonstrate colocalization (yellow) of SP-A with clathrin, type II cells were incubated with FITC-labeled SP-A (green) for 3 min at 37°C and subsequently with anti-clathrin antibody and cyanine-3-labeled anti-mouse IgG (red). Bottom left: colocalization (yellow) of FITC-labeled SP-A (green) internalized for 3 min and an antibody against the early endosomal marker EEA1 (red). Bottom right: potassium-depleted type II cells were incubated with 1 mg/ml of Lucifer yellow for 15 min at 37°C and then with tetramethylrhodamine isothiocyanate-labeled Maclura pomifera lectin, demonstrating active fluid-phase endocytosis under potassium-depleted conditions. From Stevens PA, Wissel H, Zastrow S, Sieger D, and Zimmer K-P. Surfactant protein A and lipid are internalized via the coated-pit pathway by type II pneumocytes. Am J Physiol Lung Cell Mol Physiol 280: L141-L151, 2001.
